What was measured
Serum carnosinase (CN1) activity, protein concentration and ELISA reactivity in children and adults, and kinetics of carnosine hydrolysis in the presence of homocarnosine.

What the authors reported

CN1 activity varied widely between individuals and rose with age, yet protein concentrations did not differ between children and adults; sera from children but not adults were ELISA positive and DTT raised adult signals, indicating different allosteric conformations. Homocarnosine inhibited CN1 dose-dependently: 80 microM lowered Vmax for carnosine from 440 to 356 pmol/min/microg and raised Km from 175 to 210 microM (Ki about 240 microM). The authors suggest high CSF homocarnosine explains slower carnosine degradation in CSF.

Limits of this study

Enzyme studies on human sera; no clinical outcomes. Abstract only.
